| 概要 | Environmental DNA (eDNA) metabarcoding is increasingly used to monitor marine biodiversity, but benthic organisms have rarely been targeted, especially in Japan. We collected 20 L of surface and bottom seawater from depths of 17 m and 34 m in Ise Bay, central Japan, and analyzed brittle star eDNA using the mitochondrial 16S rRNA primer set 16SOph1. Metabarcoding detected five brittle star species, whereas direct collection detected only two. DNA from bottom-dwelling species was also detected in surface water. These results indicate that brittle star metabarcoding is feasible at depths of several tens of meters and that benthic eDNA can be transported vertically to surface waters. |
